细胞外酸化速率ECAR探针
细胞外ECAR纳米探针是一种被充分证明了不会穿透细胞膜进入细胞的纳米探针,探针具有优异的水中分散性和生物相容性,能在不影响细胞正常生理代谢的情况下,通过磷光寿命测量实现细胞外的pH值或酸化程度的实时测量。探针在不同的pH时可呈现出不同的颜色,用户可以简便地通过颜色变化来判断细胞培养基的pH变化。探针的磷光寿命随着pH变化非常明显。探针长达毫秒ms级的磷光寿命,使得测量的准确度和分辨率大幅度提高,试剂的测量精度高达0.005 pH变化,非常适用于细胞外环境微小pH变化的测量。
该试剂具有优异的稳定性,在37度细胞培养环境中,其磷光寿命在连续测量180分钟时,均未出现明显的波动,表明试剂具有良好的测量稳定性和可靠性。通过连续不断地测量细胞外pH的变化,可以通过pH变化速率计算出细胞外酸化速率(ExtraCellular Acidificatioin Rate, ECAR)值,实现细胞能量代谢水平,尤其是经过糖酵解途径的能量代谢水平的定量评估。
该试剂可以在96孔板中实现活细胞ECAR水平的测量,无需采购昂贵的Seahorse或者O2K设备和耗材进行测量,客户只需配备带有荧光检测功能、最好是时间分辨荧光(time-resolved fluorescence)功能的酶标仪,就可以实现ECAR的高通量测量,大大节约了测量成本。
The extracellular acidification rate (ECAR) nanoprobe is a nanomaterial that has been thoroughly proven not to penetrate cell membranes. It exhibits excellent water dispersibility and biocompatibility, allowing for real-time measurement of extracellular pH or acidification levels via phosphorescence lifetime measurements, the nanoprobe do not interfere normal cellular physiological metabolism. The nanoprobe exhibits different colors at different pH values, enabling users to easily distinguish pH changes in the culture media through color variations. The phosphorescence lifetime of the nanoprobe varies significantly with pH changes. The nanoprobe's long phosphorescence lifetime, (upto millisecond level), significantly improves measurement accuracy and resolution. The reagent has a measurement precision as high as 0.005 pH units, making it highly suitable for measuring subtle pH changes in the extracellular environment.
This reagent exhibits excellent photostability. In a 37°C cell culture environment, its phosphorescence lifetime remains remarkably stable over continuous measurements for 180 minutes, indicating excellent measurement stability and reliability. By continuously measuring extracellular pH changes, the extracellular acidification rate (ECAR) can be calculated based on the rate of pH change, enabling quantitative evaluation of cellular energy metabolism levels, especially those involving the glycolytic pathway.
This reagent can be used to measure the ECAR levels of live cells in 96-well plates. There is no need to purchase expensive Seahorse or O2K equipment and consumables. Customers only need a microplate reader equipped with fluorescence detection capabilities, preferably time-resolved fluorescence, to achieve high-throughput ECAR measurements, significantly reducing measurement costs.

The extracellular acidification rate (ECAR) reagent developed by Fluorotool Biotech has an exceptionally large Stokes shift (~280 nm), effectively minimizing interference from endogenous bioluminescence. With a phosphorescent lifetime extending to the millisecond range, the reagent can be further enhanced for bioluminescence resistance through simple time-resolved fluorescence measurement techniques.
